丹参酮ⅱa对血管平滑肌细胞周期调控因子cyclind1 和 cdk4 表达的影响

【摘要】 摘要:目的:研究丹参酮ⅱa 拮抗同型半胱氨酸诱导的增殖血管平滑肌细胞周期的调控机制。方法:建立 h cy 诱导的兔vsm c 增殖模型,与不同浓度的丹参酮ⅱa 共同培养 48 h 后,用流式细胞仪分析 vsm c 细胞周期;检测细胞周期调控因子 cy-clind1 和 cdk4 的表达。结果:0.25 m g l 1、0.50 m g l 1、1.00 m g l 1丹参酮ⅱa 组 s 期细胞数明显减少,g0/g1期细胞数明显增多,与 h cy 组比较,差异有统计学意义(p < 0.01);cyclind1 和 cdk4 的表达量均随丹参酮ⅱa 剂量增加而减少。结论:丹参酮ⅱa 拮抗 h cy 诱导的兔 vsm c 增殖,其主要机制可能与 cyclind1 及 cdk4 的低表达所致的细胞周期抑制有关。a bstract:objective:to study the m echanism oftanshinoneⅱa in the regulation ofvascular sm ooth m uscle cellinduced by hom ocys-teine.m ethods:establish vsm c m ultiplicative m odelofrabbits induced by h cy.use flow cytom eterto analyze vsm c m itotic cycle af-ter48h with differentconcentration oftanshinone ⅱa.detectregulating factor cyclind1 and cdk4 expression ofm itotic cycle.rre-sults:the s phase cellnum bersignificantly reduced and g0/g1phase cellnum ber obviously increased in 0.25 m g l 1,0.5 m gl 1,1 m g l 1tanshinoneⅱa group.the difference was significantcom pared with h cy group(p < 0.01).the cyclind1 and cdk4expression am ountdecreased following the increase of tanshinoneⅱa.conclusion:the m ain m echanism of tanshinoneⅱa againstvsm c proliferation induced by h cy is possible referring to m itotic cycle inhibition caused by cyclind1 and cdk4 low expression
